Photosystem I catalyzes the light-driven transfer of electrons from plastocyanin or cytochrome c6 located in the lumen to ferredoxin or flavodoxin located in the stroma. This chapter focuses on electron transfer within Photosystem I, with special emphasis on the kinetic and thermodynamic parameters of the different electron transfer steps and on the spectroscopic characterization of the Photosystem I electron-transfer cofactors. The reduction of ferredoxin and flavodoxin by Photosystem I is also briefly reviewed, together with some functional and structural aspects of NADP+ photoreduction.
