Multi-compound treatment has attracted increasing interest in recent years due to vast unexplored prospects for novel therapeutics. In this chapter, we discuss the experimental design and statistical analysis of compound synergy assays. In particular, we describe how acoustic dispensing enables flexible layouts of assay plates, which greatly simplifies the study of compound combinations, and discuss measures of synergy as well as statistical properties when resampling based statistics are employed.
