This chapter deals firstly with the thermodynamics of the titration process in isothermal titration calorimetry. As the motivation for this topic we start studying in detail the Wiseman isotherm which describes the binding to a macromolecule with a single ligand binding site. After a detailed scrutiny, the Wiseman isotherm appears as a case where one error cancels the effect of another error. This is because the thermodynamics of the titration process was not carefully taken into consideration in the construction of the model. To avoid this problem in the future, we propose the present approach where the first step is the study of the thermodynamic models of the titration process from a rigorous point of view.
