During the last two decades NMR methods have been rapidly developed to study proteins in living Escherichia coli cells. This chapter reviews the labeling strategies and sample preparations for protein NMR in E. coli and summarizes some of the key advances including the determination of protein high-resolution 3D structures and the characterization of protein interactions and stabilities in E. coli cells. In addition, the limitations and future applications are discussed.
