This chapter summarises some of the techniques used in the discovery and development of small molecule protein–protein interaction (PPI) inhibitors. A description of the key considerations is supported by a selection of case studies from the literature exploring different PPI targets and a range of hit-finding techniques. A table of selected examples of PPI inhibitors that have progressed into clinical trials is also given.
